(Part 2) Molecular characterization of Mycobacterium ulcerans DNA gyrase and identification of mutations reduced susceptibility to quinolones in vitro

چکیده

Buruli ulcer disease is a neglected necrotizing and disabling cutaneous tropical illness caused by Mycobacterium ulcerans (Mul). Fluoroquinolone (FQ), used in the treatment of this disease, has been known to act inhibiting enzymatic activities DNA gyrase; however, detailed molecular basis these characteristics FQ resistance mechanisms Mul remains unknown. In study investigated mechanism gyrase contribution vitro using recombinant proteins from subsp. shinshuense Agy99 strains with reduced sensitivity FQs. Furthermore, interaction between amino acid residues WT/mutant side chains was assessed via docking studies. This first elaborative demonstrating mutations GyrA subunit vitro.

Inhibitory activities of quinolones against DNA gyrase of Chlamydia pneumoniae.

The gyrA and gyrB genes of Chlamydia pneumoniae TW-183 were cloned, and their proteins were purified by use of a fusion system with a maltose-binding protein. The 50% inhibitory concentrations of garenoxacin, sparfloxacin, moxifloxacin, gatifloxacin, and levofloxacin were 10.1, 47.5, 39.6, 64.2, and 156.9 microg/ml, respectively, and the MICs against C. pneumoniae TW-183 were 0.008, 0.016, 0.06...

Study of Mutations in the DNA gyrase gyrA Gene of Escherichia coli

Quinolones are a large and widely consumed class of synthetic drugs. Expanded-spectrum quinolones, like ciprofloxacin are highly effective against Gram-negative bacteria, especially Escherichia coli. In E. coli the major target for quinolones is DNA gyrase. This enzyme is composed of two subunits, GyrA and GyrB encoding by gyrA and gyrB, respectively. Mutations in either of these genes cause qu...

DNA gyrase, topoisomerase IV, and the 4-quinolones.

For many years, DNA gyrase was thought to be responsible both for unlinking replicated daughter chromosomes and for controlling negative superhelical tension in bacterial DNA. However, in 1990 a homolog of gyrase, topoisomerase IV, that had a potent decatenating activity was discovered. It is now clear that topoisomerase IV, rather than gyrase, is responsible for decatenation of interlinked chr...
